Won't start unless gas in cylinders

Yami 650 93 Okay the engine will only run if you put gas directly into the clyinders??? The engine will run great all day once that is done..i rebuilt the carb,changed the choke to a bulb type primer, i also added a in-line filter in the line...I really need help?? If you guys think the jets need tuning please walk me thru it because i run premix(don't wanna lean it out) any ideas
