I have a 56" inflatable o-brien tow-tube, with the tube itself being made of Vinyl. (the tube sits inside the cloth bag with attached rope)

Somehow the4 vinyl tube itself got a pinky sized hole cut into the side of it. (I think I dragged it across a nail or something)

What is the best heavy duty patch, for the money, to use to patch it?

I just want a patch that is going to be permanent and last, considering that I commonly have 200# people continually bouncing 2 feet in the air on it.

I bought something called "tear-aid type B" from west-marine, but it looks really flimsy, and I don't want to even open it If its going to leak the second a big fat guy (or girl) gets on the tube.

Go to a swimming pool store and get a patch from them. They deal in heavy vinyl's for pool liners.
