Hey so I got a 1995 four Winns 190 horizon I ended up using g the ski tow hook for towing a two person tube and after getting the boat home and inspecting I saw that the little screw that stops the ski hook From turning/spinning was lose and won’t catch. I ended up re tightening the backing plate/ main bolt but was wondering if I should use a bigger screw & if I should should seal it ? If so how would I go about re sealing it or making it leak proof?

yes, you need to re-seal. I suggest 3M 4200 UV

and yes, on that style of tow hook, there is the screw you mentioned. pull the tow hook off or rotate it. since giong the a larger screw may require you to drill a larger hole in the escutcheon plate, I would fill the hole with marine tex epoxy, then let gently put your screw in. the marine-tex will help retain the screw

For towing a tube, especially a multiple rider tube, you might consider using the transom tie-down u-bolts and a bridle. The tow hook may or may not stand up to lots of tube-towing, but you likely will see lots of wear on the eyeloop of your tow rope. A bridle connects to the very strong transom tie-downs and lessens the wear on your tow rope.
