sand after you prime?

I am painting a portion of my boat above the rub rail. I have already bought the primer and paint also sanded the portion to be painted. This weekend I am going to get at least the primer down and if possible the first coat. After I put the primer down, do I lightly sand then paint?

I always do, 320 grit or sometimes just a 3M Scotch Brite pad. Knocks the nubs down and scuffs it a bit to give the next coat something to Bite in to.
