Is it necessary to have the quick disconnect fittings on both sides of the main fuel line?

no, built in tanks only have the motor connection. or if you have portable tanks, the tank end is ok.

I have portable tanks and there were the connections on both sides but fit loosely and leaked, so I went to NAPA and got some fittings to connect it all together. I wont have any problems will I?

Actually it depends on the motor. You don't "need" a connector on either end. Some tanks are set up for a quick connect, some have the hose fitting. On V4 loopers and all V6 & V8's the quick connect fitting is viwed as a fuel restriction. On smaller motors it's still a restriction but the motor still gets enough fuel.
