Should I have telltale when cold? 2003 Yamaha 4 stroke f15

Running in bucket. Should telltale start right away?. I only ran it for 15 seconds.

Yes it should, might blow some compressed air into tell tail to see if it is plugged

Impeller was gone. Too long in Arizona I guess. Do you think running it for 30 seconds a few times hurt it? The engine didn't even get warm.

running without water damages the impellar, 30 seconds is long enough to get engine hot, you'll be fine
