My houseboat has a SINGLE Volvo aq270 duoprop drive.
Sitting at the dock, it will shift into forward or reverse.
After running for a few minutes, it will not go into reverse.

Can this be caused by low oil in the drive?

Unfortunately, to service the drive I must have the boat taken out of the water, at considerable cost, so I want to know ahead of this is this is likely to be the cause

I would think you are going to need to pull the boat out and check the drive. When was the last time the oil was changed/checked in the drive?

You can potentially check the shift adjustment while it is in the water of course you will have to put on your snorkelling gear to do it :) The easiest way may be to disconnect the cable at the console shifter, take the cover off the back of the drive and make sure the drive is in its neutral detent and make the adjustment up at the console so that it is definitley in the center of neutral.
