1972 mercury 500 50 timing

hi every one ,need some help timing my mercury 500 50,fly wheel has two timing marks,the first one i line upto the tdc on the timing decoil, which makes the second mark line up to the dist,so both marks on fly wheel are now alined, it should be timed at 35 degress btdc.is this with throttle fully open or in neutral also which mark on the flywheel do i take notice of when moving the flywheel to the 35 degress mark .any advice would be really greatfull.
